Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Published work supports the view that engaging two incretin receptors produces changes in glucose handling and body weight larger than those seen with single-receptor activation. Why that difference arises is not fully settled. Open questions include how much of the observed weight effect depends on central versus peripheral signaling, and whether the two receptors form interacting complexes. Most reported findings come from controlled trials and animal models, and translation between species is imperfect. Further research is expected to refine these points over time.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.

The compound first appeared in the scientific literature as an investigational agent for type 2 diabetes. Clinical development proceeded through phase 1, phase 2, and phase 3 programs that measured glycemic control as a primary endpoint while recording body weight as a secondary outcome. Regulatory approval in the United States followed in 2022 for glycemic control, and a separate indication for chronic weight management was added later. Subsequent trials have examined cardiovascular outcomes in adults with elevated cardiovascular risk. Debates continue over how much of the observed effect derives from each receptor arm.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.
Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
=== In the nervous system === In addition to roles in mediating cell survival, studies by Mark Mattson and others have shown that NF-κB has diverse functions in the nervous system including roles in plasticity, learning, and memory. In addition to stimuli that activate NF-κB in other tissues, NF-κB in the nervous system can be activated by Growth Factors (BDNF, NGF) and synaptic transmission such as glutamate. These activators of NF-κB in the nervous system all converge upon the IKK complex and the canonical pathway. Recently there has been a great deal of interest in the role of NF-κB in the nervous system. Current studies suggest that NF-κB is important for learning and memory in multiple organisms including crabs, fruit flies, and mice. NF-κB may regulate learning and memory in part by modulating synaptic plasticity, synapse function, as well as by regulating the growth of dendrites and dendritic spines. Genes that have NF-κB binding sites are shown to have increased expression following learning, suggesting that the transcriptional targets of NF-κB in the nervous system are important for plasticity. Many NF-κB target genes that may be important for plasticity and learning include growth factors (BDNF, NGF) cytokines (TNF-alpha, TNFR) and kinases (PKAc). Despite the functional evidence for a role for Rel-family transcription factors in the nervous system, it is still not clear that the neurological effects of NF-κB reflect transcriptional activation in neurons.
==== Reservation reconstituted ==== On July 9, 2020, the United States Supreme Court ruled in a 5–4 decision that the original treaties, and promise of a reservation, with the Five Civilized Tribes (specifically the Muscogee in McGirt v. Oklahoma) were never withdrawn. This decision allowed for the restoration of the reservation status of the Cherokee Nation in a later decision. The majority opinion was held by justices Sonia Sotomayor, Ruth Bader Ginsburg, Elena Kagan, Stephen Breyer, and Neil Gorsuch. Hogner v. Oklahoma was decided on March 11, 2021, in Oklahoma Courts and found that the Cherokee Nation "reservation was established and had never been disestablished." Cherokee Nation Principal Chief Chuck Hoskin, Jr. said: "We have long held that Cherokee Nation has a reservation, rooted in our treaties, as the Supreme Court of the United States has now affirmed" and "This proposed legislation will cement our reservation boundaries and the broad tribal jurisdiction the Supreme Court recognized in the McGirt decision. We will continue to work with the state of Oklahoma and our federal partners to ensure the safety of the public." The United Keetoowah Band of Cherokee Indians claims it has rights to the reservation as well as a "successor in interest" because they contend the original Cherokee Nation was "sunset" after "the last of its original members who signed the Dawes Roll died in 2012 at age 107." Therefore, the current Cherokee Nation is no longer the same Cherokee Nation that made the agreements with the federal government, it is only a "successor" like the UKB.
=== Amphiphilic FSL Kode construct === The FSL Kode construct by nature of its composition in possessing both hydrophobic and hydrophilic regions are amphiphilic (or amphipathic). This characteristic determines the way in which the construct will interact with surfaces. When present in a solution they may form simple micelles or adopt more complex bilayer structures with two simplistic examples shown in Fig. 5a. More complex structures are expected. The actual nature of FSL micelles has not been determined. However, based on normal structural function of micelles, it is expected that it will be determined in part by the combination of functional group, spacer and lipid together with temperature, concentration, size and hydrophobicity/hydrophilicity for each FSL Kode construct type. Surface coatings will occur via two theoretical mechanisms, the first being direct hydrophobic interaction of the lipid tail with a hydrophobic surface resulting in a monolayer of FSL at the surface (Fig. 5b). Hydrophobic binding of the FSL will be via its hydrophobic lipid tail interacting directly with the hydrophobic (lipophilic) surface. The second surface coating will be through the formation of bilayers as the lipid tail is unable to react with the hydrophilic surface. In this case the lipids will induce the formation of a bilayer, the surface of which will be hydrophilic. This hydrophilic membrane will then interact directly with the hydrophilic surface and will probably encapsulate fibres.

Radiation therapy is usually reserved for people who have tumours remaining after surgery. These people often also receive medication to lower GH levels. Radiation therapy is given in divided doses over four to six weeks. This treatment lowers GH levels by about 50 percent over 2 to 5 years. People monitored for more than 5 years show significant further improvement. Radiation therapy causes a gradual loss of production of other pituitary hormones with time. Loss of vision and brain injury, which have been reported, are very rare complications of radiation treatments.

Ion-exchange membranes are traditionally used in electrodialysis or diffusion dialysis by means of an electrical potential or concentration gradient, respectively, to selectively transport cationic and anionic species. When applied in an electrodialysis desalination process, anion- and cation-exchange membranes are typically arranged in an alternating pattern between two electrodes (an anode and a cathode) within the electrodialysis stack. A galvanic potential is supplied as a voltage generated at the electrodes. A typical industrial electrodialysis stack consists of two chambers: a product-water chamber and a concentrate-reject chamber. During stack operation, salts are transferred from the product to the concentrate. As a result, the reject stream is concentrated up while the product stream is desalted. Exemplary applications of ion-exchange membranes utilized in electrodialysis and EDR include seawater desalination, industrial wastewater treatment of highly scaling waters, food and beverage production, and other industrial wastewaters. RVT-802 is an investigational treatment for congenital athymia, primarily associated with DiGeorge syndrome. It is a tissue-based therapy that consists of cultured donor thymus-derived tissue. RVT-802 consists of donor thymus-derived tissue that is cultured and surgically implanted into the recipient In patients with congenital athymia, the thymus gland is absent. Because of the crucial role the thymus gland plays in the maturation and differentiation of T cells, athymia results in severe immunodeficiency, typically resulting in death within the first two years of life. RVT-802 is manufactured by extracting thymus tissue from infants undergoing cardiac surgery, depleting it of immature T cells to prevent graft-versus-host disease, then implanting the processed tissue into the recipient's leg, where it fulfils the immunological role of the thymus.
A flagellate is a cell or organism with one or more whip-like appendages called flagella. The word flagellate also describes a particular construction (or level of organization) characteristic of many prokaryotes and eukaryotes and their means of motion. The term presently does not imply any specific relationship or classification of the organisms that possess flagella. However, several derivations of the term "flagellate" (such as "dinoflagellate" and "choanoflagellate") are more formally characterized.
In a typical nuclear reactor fueled with uranium-235, the presence of 135Xe as a fission product presents designers and operators with problems due to its large neutron cross section for absorption. Because absorbing neutrons can impair a nuclear reactor's ability to increase power, reactors are designed to mitigate this effect and operators are trained to anticipate and react to these transients. This practice dates to the first fission piles, constructed by the Manhattan Project during the Second World War. Enrico Fermi suspected that 135Xe would act as a powerful neutron poison and followed the advice of Emilio Segrè by contacting his student Chien-Shiung Wu. Wu's unpublished paper on 135Xe verified Fermi's guess that it absorbed neutrons and was the cause of the disruptions to the B Reactor then in use at Hanford, Washington to breed plutonium for the American implosion bomb. During periods of steady state operation at a constant neutron flux level, the 135Xe concentration builds up to its equilibrium value for that reactor power in about 40 to 50 hours. When the reactor power is increased, 135Xe concentration initially decreases because the burn up is increased at the new higher power level. Because 95% of the 135Xe production is from decay of 135I, which has a 6.58 hour half-life, the production of 135Xe remains constant; at this point, the 135Xe concentration reaches a minimum. The concentration then increases to the new equilibrium level (more accurately steady state level) for the new power level in roughly 40 to 50 hours.

Some publications write the nucleotides in a different order (e.g., some authors choose to group two purines together and the two pyrimidines together; see also models of DNA evolution). These differences in notation make it important to be clear regarding the order of the states when writing the
On the morning of 8 September, representatives of the Fatherland Front - Kimon Georgiev, Nikola Petkov, Dimitar Neykov, Kiril Dramaliev and Dimo Kazasov - met with the Prime Minister, protesting the dispersal of opposition demonstrations in the previous days and demanding that rallies be allowed in the major cities. Kimon Georgiev hosted a meeting of the Fatherland Front's National Committee at 4 p.m. on 8 September. The government's composition was settled upon, and its policy text is approved. The composition of the future government and the new regents were specified at a meeting between Kimon Georgiev, Dobri Terpeshev, Nikola Petkov and Damyan Velchev at Georgiev's home at 4 pm on 8 September. It was agreed that the cabinet would include four representatives each of the BRP, Zveno and BZNS-Pladne, two of the BRSD and two independents, and that the prime minister would be Kimon Georgiev, a decision agreed with Soviet dictator Joseph Stalin. Georgiev spent the night of the coup with Damyan Velchev, Nikola Petkov, and Traicho Dobroslavsky at the home of Yanko Antonov near the Eagles Bridge - he was a neighbor of Peter Vranchev, in whose apartment the Communist leaders - Dobri Terpeshev, Anton Yugov, Georgi Chankov, Angel Tsanev, and Katya Avramova were at the time. The coup began at 2 a.m. on 9 September with the seizure of the War Ministry building. War Minister Ivan Marinov sided with the coup and issued the appropriate orders to the First Infantry Division and the School for Reserve Officers.
== Structure == The human OGT gene has 1046 amino acid residues, and is a heterotrimer consisting of two 110 kDa subunits and one 78 kDa subunit. The 110 kDa subunit contains 13 tetratricopeptide repeats (TPRs); the 13th repeat is truncated. These subunits are dimerized by TPR repeats 6 and 7. OGT is highly expressed in the pancreas and also expressed in the heart, brain, skeletal muscle, and the placenta. There have been trace amounts found in the lung and the liver. The binding sites have been determined for the 110 kDa subunit. It has 3 binding sites at amino acid residues 849, 852, and 935. The probable active site is at residue 508. The crystal structure of O-GlcNAc transferase has not been well studied, but the structure of a binary complex with UDP and a ternary complex with UDP and a peptide substrate has been researched. The OGT-UDP complex contains three domains in its catalytic region: the amino (N)-terminal domain, the carboxy (C)-terminal domain, and the intervening domain (Int-D). The catalytic region is linked to TPR repeats by a translational helix (H3), which loops from the C-cat domain to the N-cat domain along the upper surface of the catalytic region. The OGT-UDP-peptide complex has a larger space between the TPR domain and the catalytic region than the OGT-UDP complex. The CKII peptide, which contains three serine residues and a threonine residue, binds in this space.
Despite his triumph in Rome, he lost his opening round match at the French Open. During the grass court season, Zverev made another singles final and again faced Federer at the Halle Open, but could not defeat him this time. He and his brother also finished runner-up in the doubles event to Łukasz Kubot and Marcelo Melo. At Wimbledon, he achieved his best result at a major event to date, ultimately losing in the fourth round to the previous year's runner-up Milos Raonic in a tight five-set match. Back on hard courts, Zverev won his last two titles of the season in August. He won the Washington Open as well as a second consecutive Masters title at the Canadian Open, only dropping a single set at each tournament in each of his opening matches. Notably, he needed to save three match points in his first match in Canada against Richard Gasquet, including a 49 shot rally. He then defeated Roger Federer in the final to become the first player outside of the Big Four to win multiple Masters titles in the same season since David Nalbandian in 2007. Despite this success, he was upset in the second round of the US Open by fellow Next Gen player Borna Ćorić. At the end of the season, Zverev qualified for both the inaugural Next Generation Finals as one of the top seven 21-and-under players, and the ATP Finals as one of the top eight players in the world. He opted to skip the former event to focus on the latter. At the ATP Finals, Zverev was grouped with Roger Federer, Marin Čilić, and Jack Sock.
From the previous boundary changes in 2013 the council comprised 126 councillors representing 63 electoral divisions, with each division electing one, two or three councillors. Elections are held every four years. New division boundaries were drawn up to take effect from the 2025 election, reducing the number of councillors to 98.

It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
A fatty diacid side chain promotes reversible binding to serum albumin. This association slows renal clearance and protects the peptide from rapid enzymatic degradation. The result is a prolonged circulation time that supports weekly administration.
The first regulatory approval, for type 2 diabetes, was granted in the United States in 2022. An additional approval for chronic weight management followed in 2023. Availability and approved indications vary by country and are set by each national regulator.
It is a synthetic peptide and a dual agonist of two incretin receptors. It is not a small molecule, and it is not structurally related to the older single-receptor peptide agonists.
