This is a working overview of GIP receptor, written for readers who want more than a one-paragraph summary but less than a textbook.

Tirzepatide is a synthetic peptide developed as a dual agonist at the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors. Its structure is built on a GIP-derived backbone with non-natural amino acid substitutions and a fatty diacid side chain that promotes albumin binding and slows clearance. That modification supports once-weekly subcutaneous dosing. Registrational trial programs reported reductions in body weight and glycated hemoglobin alongside the drug's glycemic effects.
Both receptors are class B G protein-coupled receptors that signal largely through Gs-mediated cyclic AMP production. Activation within pancreatic islets increases glucose-dependent insulin secretion and suppresses glucagon release when glucose is elevated. Outside the pancreas, signaling in the central nervous system and gut appears to influence appetite and gastric emptying. The relative contribution of each receptor to observed clinical effects remains under investigation, and the two pathways are not simply additive in practice.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
| Property | Value | Notes |
|---|---|---|
| Molecular class | Synthetic 39-residue peptide | GIP-derived backbone with non-natural residues |
| Molar mass | Approximately 4.8 kDa | Peptide chain plus linker and lipid modifications |
| Appearance | White to off-white powder | Typical of lyophilized research material |
| Solubility | Soluble in water and aqueous buffer | Practically insoluble in nonpolar solvents |
| Storage temperature | Typically −20 °C for dry powder | Solutions usually held at 2–8 °C short term |
Routine characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity assessment, usually with ultraviolet detection near 214 nanometers. Intact mass measurement by liquid chromatography coupled to mass spectrometry confirms molecular identity against a theoretical value. Sequence-level confirmation uses enzymatic digestion followed by tandem mass spectrometry, an approach known as peptide mapping. Amino acid analysis gives an independent check on composition. Circular dichroism spectra are used to estimate helical content in aqueous buffer.
Stability depends strongly on physical form. The dry powder is generally regarded as stable for extended periods when held at or below minus twenty degrees Celsius in a sealed, desiccated container. In solution, degradation pathways include deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation. Reaction rates for these pathways rise with temperature. Repeated freezing and thawing of solutions promotes aggregation, and light exposure can accelerate some oxidative changes. Buffer composition and pH influence which pathway dominates at a given temperature.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
need not be constant – in general, they depend on two thermodynamics variables if the fluid contains a single chemical species, say for example, pressure and temperature. Any equation that makes explicit one of these transport coefficient in the conservation variables is called an equation of state. Apart from its dependence of pressure and temperature, the second viscosity coefficient also depends on the process, that is to say, the second viscosity coefficient is not just a material property. Example: in the case of a sound wave with a definitive frequency that alternatively compresses and expands a fluid element, the second viscosity coefficient depends on the frequency of the wave. This dependence is called the dispersion. In some cases, the second viscosity
Changes in nearby tissue is not the only possible impact of ligand therapy, there may be immunologic responses from the target tissue that cause changes remotely. This has been dubbed the "abscopal effect". While this mechanism is not well understood, it explains the impact of other tissue, both benign and malignant, after targeted radiotherapy.
== Clinical evidence == In a six-month randomized trial published in The New England Journal of Medicine in 2019, 168 people with type 1 diabetes were assigned either Control-IQ with a t:slim X2 and Dexcom G6 or a sensor-augmented pump. Mean time with glucose in the target range of 70–180 mg/dL rose from 61% to 71% in the Control-IQ group and remained about 59% in the control group, an adjusted difference of 11 percentage points, or 2.6 hours per day. No severe hypoglycemic events occurred; one participant using the closed-loop system developed diabetic ketoacidosis after an infusion-set failure. The trial was funded by the National Institute of Diabetes and Digestive and Kidney Diseases; Tandem supplied systems and technical support but, according to the publication, did not control the study design, conduct or analysis. A 13-week randomized trial published in the same journal in 2025 enrolled 319 adults with insulin-treated type 2 diabetes. Time in the target range increased from 48% to 64% with Control-IQ, compared with 51% to 52% in a control group that continued its previous insulin method; the adjusted difference was 14 percentage points. Glycated hemoglobin fell by 0.9 percentage points in the automated-delivery group and by 0.4 points in the control group. One severe hypoglycemic event occurred in the automated-delivery group. The study was funded by Tandem.

=== Student Union Memorial Center === The Student Union Memorial Center, on the north side of the Mall east of Old Main, was completely reconstructed between 2000 and 2003. It replaced a 270,000-square-foot (25,000 m2) building that originally opened in 1951 and had been expanded during the 1960s and early 1970s. Designed to resemble the USS Arizona (BB-39), the building includes memorial features honoring members of the university community and others who served in the military. Memorial sculptures throughout the complex incorporate elements such as the sounds of chiming dog tags and refracted light. One of the center's best known features is the USS Arizona bell, which is housed in the building's clock tower. The 1,820-pound (826 kg) bell was one of two recovered from the battleship after the attack on Pearl Harbor and arrived on campus in July 1946. For decades, the bell was rung seven times on the third Wednesday of every month at 12:07 p.m., marking the time of the USS Arizona's sinking on December 7, 1941. The bell was also rung after Arizona Wildcats football home victories, except against other Arizona schools. In December 2020, the university ended the long-standing ringing tradition after the U.S. Navy, which still owns the bell, requested that the historic object be protected from further wear.

Their southernmost settlements being around Schwansen, Hedeby and Husum, mirroring the same southern border as their Angle predecessors. As raiding was a frequent practice among the Danes, Saxons, and Wagrians, the borderlands was a hostile and unsafe area to inhabit. In response to these threats, the Danes began constructing the Danevirke. Originally built as a dyke, it was gradually expanded into a 30-kilometer earthwork with a palisade fortification, forming a barrier between the Danes and their southern neighbors. The Danevirke was strategically positioned at the narrowest point of the peninsula, with its eastern end beginning at the Treene River and extending to the Schlei Bay. This fortification served to deter Saxon and Wagrian raids while enabling the Danes to launch their own raids into southern territories. It would remain in use until 1864, being expanded and adapted to the changing military needs of the Danes multiple times. The establishment of the Danevirke not only helped to prevent Saxon and Wagrian raids into Danish territory but also served as a toll station. Danish chieftains would collect fees from traders, merchants, and peasants traveling along the Hærvejen (Heerweg/Armyway), the main trade route running through the peninsula. Saxons, Wagrians, and Danes alike used the Hærvejen to trade goods such as honey, furs, amber, glass, metalwork, and other commodities, such as livestock, with cattle and oxen being particularly important.
In two minor league seasons from 1989 to 1990, he had managed six home runs in 932 at bats–a ratio of 155 at bats per home run (AB/HR). With 15 home runs in his first year in Houston, that average shrunk to 36.9. He also exhibited extraordinary plate discipline for a rookie: while ranking tenth in the league in walks with 75, his OBP placed fifth at .387. Of the power surge, commented Bagwell to hitting coach Rudy Jaramillo, "That's awesome", to which he reacted, "Awesome? We can get more out of you than that." By altering an approach to contact the pitch with topspin as he did when he arriving to Houston, Jaramillo taught Bagwell to hit with backspin, resulting in a soaring trajectory rather than nose diving. He also habituated Bagwell to manipulate the count, waiting for a pitch to drive instead of indiscriminately swinging at any pitch that appeared to be a strike. Accounted Bagwell of the newfound advantage, "I didn't hit many home runs in the minor leagues, but when I hit one, when I got backspin on it, it went a long way." Although firmly established as the Astros' first baseman from Opening Day in 1991, Bagwell remarked years later that transitioning from third base was not automatic. First basemen approach fielding plays from their right side, which is opposite to playing third base. He recounted one conversation that occurred during an Astros pitching change in a game against St. Louis. Shortstop Ozzie Smith was on first and asked Bagwell, "How's it going?" Bagwell responded, "I'm really struggling with my backhand." Smith replied, "Well, here's what you do.
For centuries, naturalists treated lichens as self-contained, plant-like organisms distinguished only by outward appearance. Theophrastus (c. 300 BC) introduced the word lichen for crusty bark growths, yet offered little insight beyond the name. Until the mid-1700s, taxonomists lumped lichens with algae, mosses, or fungi in broad, pre-evolutionary schemes. A pivotal step came in 1700, when the French botanist Joseph Pitton de Tournefort erected the genus Lichen, acknowledging the group's distinctiveness—even while keeping it beside mosses and liverworts. Robert Morison's 1699 Herbarium, for instance, split lichens into five "Muscofungi" types, a purely morphological scheme that left little mark on later work. The Italian polymath Pier Antonio Micheli published the first recognizable lichen classification in his 1729 Nova plantarum genera. While he kept all species in the catch‑all genus Lichen—echoing Tournefort—Micheli organized them into several "orders" based on thallus texture and fruiting body form. Those informal groupings later became the nuclei of modern genera, and his morphological terminology laid the groundwork for subsequent binomial treatments. In 1753 Carl Linnaeus introduced the first coherent plant-classification scheme in Species Plantarum. He listed roughly 80 lichen species, grouping almost all under the single genus Lichen. Such compression mirrored 18th-century ignorance of lichen diversity: 'lichen' was little more than a catch-all for crusty or leafy growths on bark and stone.
A skeletal survey is useful to confirm the diagnosis of achondroplasia. The skull is large, with a narrow foramen magnum, and relatively small skull base. The vertebral bodies are short and flattened with relatively large intervertebral disk height, and there is congenitally narrowed spinal canal. The iliac wings are small and squared, with a narrow sciatic notch and horizontal acetabular roof. The tubular bones are short and thick with metaphyseal cupping and flaring and irregular growth plates. Fibular overgrowth is present. The hand is broad with short metacarpals and phalanges, and a trident configuration. The ribs are short with cupped anterior ends. If the radiographic features are not classic, a search for a different diagnosis should be entertained. Because of the extremely deformed bone structure, people with achondroplasia are often "double jointed". The diagnosis can be made by fetal ultrasound by progressive discordance between the short femur length and biparietal diameter by age. The trident hand configuration can be seen if the fingers are fully extended. Another common characteristic of the syndrome is thoracolumbar gibbus in infancy.

It acts as an agonist at both the GIP and GLP-1 receptors, two related class B G protein-coupled receptors. This dual activity distinguishes it from single-receptor GLP-1 agonists. The clinical consequences of engaging both receptors are still being characterized.
It is given by subcutaneous injection, generally on a weekly schedule. Dosing usually begins low and increases in steps to limit gastrointestinal side effects. Formulated product is supplied as a ready-to-use pen or vial in most markets.
No. Receptor engagement is documented, but how central appetite circuits, gut signals, and insulinotropic effects combine is incompletely resolved. Investigators continue to separate GIP-driven from GLP-1-driven contributions in animal and human models.
It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
